Nope, it will remain unchanged. Same goes for quality and enchants. Only enhancement will reset if you replace the purple essence with the orange essence.
I have used 2 Unstable Runes. Rune is still consumed whatever the enhancement was successful or not. No rune fragment was obtained when the enhancement failed.
